Purpose of research: Applied research

The expression of a variety of detoxifying enzymes is regulated through the transcription factor Nrf2. These metabolic enzymes catalyze the conjugation and thus trigger the elimination of biologically reactive intermediates such as electrophiles to protect the skin from acute and chronic toxicity. However, this mechanism is as yet not well understood with respect to its relevance for the toxicity of consumer products. In addition, there is a great need for in vitro screening assays suited to identify classes of compounds from consumer products that may compromise this cellular defence mechanism.We are aimed at identifying chemical compounds from consumer products that interact with this Nrf2-dependent cellular defence system in human keratinocytes. To this end we designed a highly sensitive dual luminometer reporter gene assay to detect Nrf2 activation triggered by chemicals in vitro.
