Investigation of the DNA methylation of Fusarium graminearum by applying different chemical compounds

One of the reasons why the infection of grains with fusarium gets so much attention is the production of mycotoxins. This occurs all the more the fungus is put under stress, especially when there are sub-lethal concentrations of fungicides (Azols). In addition, by using a lower fungicide dose there is the possibility that resistance is built up by fusarium. To evaluate the influence of chemical substances on the methylation of genes of the mycotoxin gene cluster, the genes Tri5 (trichodien synthase) and the Azol resistance genes Cyp A, B, C were surveyed. Using the bisulfite treatment, the methylation status of cytosines can be determined. Bisulfite converts all unmethylated cytosines into finally thymines but saves the methylated cytosines. In a subsequent sequencing the methylated cytosines are unchanged and appear as cytosines. Several chemical compounds are known which influence the DNA methylation, including ammonium sulphate and folic acid. A correlation between the Azole resistance, Tri5 gene inactivation and DNA methylation will be researched and a mycotoxin and resistance reduction by externally influencing the DNA methylation be demonstrated.
