Purpose of research: Applied research

According to the EST protocol, proliferation and differentiation of embryonic stem cells (ESC) are performed in a culture medium containing 15 % - 20 % fetal calf serum. Unfortunately the quality of the serum differs from batch to batch. Therefore, different batches of serum have to be pre-tested before they can be used in the EST. In order to standardize the test system our aim is to establish serum-reduced and/or serum-free culture conditions for proliferation and differentiation of ESC. Serum-free conditions would considerably increase the level of standardization of the EST an important prerequisite for applications in High Throughput Screening Systems (HTPS).
