The aim of the project is the evaluation of a virus elimination approach by cryopreservation of strawberry. An evaluation of 42 strawberry cultivars from the germplasm collection of Julius Kühn-Institut resulted in the detection of up 4 viruses in 31 strawberry cultivars. In a parallel approach 15 strawberry cultivars were regenerated after cryopreservation and transferred to the green house. The plants were tested for the occurrence of viruses and showed a much lower infestation compared to the material before cryopreservation. Up to now, nothing is known about the role of cryopreservation or its preliminary steps like in-vitro culture, in-vitro propagation and regeneration in virus elimination of strawberry tissue. To proof the influence of these steps, virus infected strawberry cultivars will be cultured in-vitro and cryopreservated in parallel for several months. Plant material will be collected during each step and investigated for the occurrence of 9 strawberry viruses (Strawberry vein banding virus (SVBV), Arabis mosaic virus (ArMV), Raspberry ringspot virus (RpRSV), Strawberry crinkle virus (SCV), Strawberry latent ringspot virus (SLRV), Strawberry mild yellow edge virus (SMYEV), Tomato black ring virus (TBRV), Tomato ringspot virus (ToRSV) and Strawberry mottle virus (SMoV)). The results contribute to the development of a virus free plant stock of strawberry genetic resources.
