The recombinant bovine somatotropin (rBST) is a synthetic peptide hormone. It will be produced and used in industrial scale in several countries in order to increased milk production at lactating cows. The application of rBST therefore may result only in economic benefits although no therapeutic indications have been considered in the target animal species to date. For this reason many countries have approved and used rBST treatment in food production, e.g. in USA it is permitted to use this treatment to increase the milk production, so that about 50% of the dairy cows are treated with rBST. According to the Commission Decision (1999/879/EG) the using of rBST for lactating cows is forbidden. In order to consumers safety it is essential to develop an effective and sensitive detection method to identify products originated from animals treated with rBST. However, up to now there is no confirmatory method to control the use of rBST in lactating cows. The methods described actually in the literature for the detection of rBST have minor specificity (e.g. immunoassay or size exclusion chromatography with UV detection). The scientific goal of the project therefore is the development of a method for the accurate and specific detection of rBST in milk, in urine and in plasma, as well as in edible tissues, based on the difference of the mass specific data between endogenous and recombined BST. The aim of the present work is the development of a reliable detection method, which is able to differentiate between the endogenous and recombined BST residues based on their mass differences using HPLC and nanospray mass spectrometry.
